This gene encodes a five transmembrane protein that functions as a major regulator of the innate immune response to viral and bacterial infections. The encoded protein is a pattern recognition receptor that detects cytosolic nucleic acids and transmits signals that activate type I interferon responses. The encoded protein has also been shown to play a role in apoptotic signaling by associating with type II major histocompatibility complex. Mutations in this gene are the cause of infantile-onset STING-associated vasculopathy. Alternate splicing results in multiple transcript variants. [provided by RefSeq, Sep 2014] Database links: Entrez Gene: 340061 Human SwissProt: Q86WV6 Human STING由379個(gè)氨基酸構(gòu)成，有數(shù)個(gè)跨膜結(jié)構(gòu)域，在多種內(nèi)皮細(xì)胞、上皮細(xì)胞中以及造血肝細(xì)胞均有表達(dá)。STING參與并促進(jìn)DNA介導(dǎo)的先天免疫反應(yīng)，即來(lái)自病毒和細(xì)菌的非CpG雙鏈DNA與cGAS蛋白結(jié)合后被催化成第二信使cGAMP；接著二聚化的STING與cGAMP結(jié)合，發(fā)生構(gòu)象變化，通過(guò)自噬小體（autophagsome）經(jīng)內(nèi)質(zhì)網(wǎng)和高爾基體進(jìn)一步進(jìn)入細(xì)胞質(zhì)，期間被泛素化后募集TBK1蛋白，再被磷酸化，之后與IRF3結(jié)合；接著IRF3被磷酸化后入核，誘導(dǎo)表達(dá)IFNs，細(xì)胞因子、T細(xì)胞募集因子等進(jìn)而增強(qiáng)機(jī)體的免疫能力，更大限度的殺傷細(xì)菌，病毒等病原體。 |
