ICOS ligand is a member of the B7 family and the immunoglobulin superfamily. Human ICOS ligand is expressed by activated monocytes/macrophages and dendritic cells. It binds to a CD28 like receptor, inducible costimulator molecule (ICOS, AILIM, CRP-1), which is expressed by activated T cells. This interaction plays an important role in the T cell costimulation pathway. Function: Ligand for the T-cell-specific cell surface receptor ICOS. Acts as a costimulatory signal for T-cell proliferation and cytokine secretion; induces also B-cell proliferation and differentiation into plasma cells. Could play an important role in mediating local tissue responses to inflammatory conditions, as well as in modulating the secondary immune response by co-stimulating memory T-cell function. Subunit: Membrane; Single-pass type I membrane protein. Subcellular Location: Isoform 1 is widely expressed (brain, heart, kidney, liver, lung, pancreas, placenta, skeletal muscle, bone marrow, colon, ovary, prostate, testis, lymph nodes, leukocytes, spleen, thymus and tonsil), while isoform 2 is detected only in lymph nodes, leukocytes and spleen. Expressed on activated monocytes and dendritic cells. Similarity: Belongs to the immunoglobulin superfamily. BTN/MOG family. Contains 1 Ig-like C2-type (immunoglobulin-like) domain. B7協(xié)同刺激分子及其相應(yīng)受體結(jié)合所產(chǎn)生的信號在調(diào)節(jié)T細(xì)胞活化及效應(yīng)性細(xì)胞因子分泌中起重要作用，具有負(fù)性調(diào)節(jié)作用的協(xié)同刺激分子對免疫應(yīng)答的適度進(jìn)行、防止自身免疫疾病的發(fā)生具有重要作用。 B7H2可協(xié)同刺激CD14 + 細(xì)胞增殖，并分泌IL-4、IL-5、IFN-β、TNF-α、GM-CSF和IL-10等細(xì)胞因子，但不能協(xié)同刺激T細(xì)胞上調(diào)IL-2的分泌。 |

Blank controlBlue):Jurkat cells (fixed with 2% paraformaldehyde (10 min)). Primary Antibody:Rabbit Anti-ICOS ligand antibody(bs-2471R), Dilution: 1μg in 100 μL 1X PBS containing 0.5% BSA;
Isotype Control Antibody: Rabbit IgG(orange),used under the same conditions );
Secondary Antibody: Goat anti-rabbit IgG-PE(white blue), Dilution: 1:200 in 1 X PBS containing 0.5% BSA.
Protocol
The cells were fixed with 2% paraformaldehyde (10 min). Primary antibody (bs-2471R, 1μg /1x10^6 cells) were incubated for 30 min on the ice, followed by 1 X PBS containing 0.5% BSA + 1 0% goat serum (15 min) to block non-specific protein-protein interactions. Then the Goat Anti-rabbit IgG/PE antibody was added into the blocking buffer mentioned above to react with the primary antibody at 1/200 dilution for 30 min on ice. Acquisition of 20,000 events was performed.
